그림 1. SPINeasy® RNA Kit for Yeast를 사용하여 Yeast (S. cerevisiae and P. pastoris) 에서 total RNA를 추출. 추출은 FastPrep®과 vortex 두가지 방법으로 진행하였으며, 추출된 샘플은 타사 X, Y, Z사의 결과와 비교하였습니다. 추출된 RNA는 Agilent® Bioanalyzer 4150 & RNA Screen tape을 사용하여 분석하였으며, RIN 과 OD를 통해 전반적인 RNA의 integrity와 purity를 확인할 수 있었습니다.

그림 2. qPCR을 통한 증폭 효율 확인. 추출된 RNA는 downstream applications과의 호환성을 확인하기 위해 qPCR을 진행하였습니다. S. cerevisiae (Figure 2A)와 P. pastoris (Figure 2B)에서 각각 추출된 RNA는 template 농도 5 log 범위로 희석되었으며, Actin에 대한 primer와 iTaq™ Universal SYBR® Green One-Step Kit RT-qPCR reagents를 사용하여 QuantStudio3 (Applied Biosystems, by ThermoScientific)장비에서 실험 진행되었습니다.
