2xYT-Broth is a nutritional rich medium providing nitrogen and growth factors optimized for the growth of recombinant Escherichia coli strains infected with the M13 bacteriophage or other filamentous single stranded DNA bacteriophages thus allowing the bacteriophage to reproduce in large quantities without exhausting the host cells.

| Preparation Method | Media Preparation: Reagents are tested and chosen for their ability to promote optimum growth of lambda phage and phagemids in E. coli when combined in 2xYT-Broth formulations. Chosen reagents are combined in the proper proportions according to the 2xYT-Broth formulation and thoroughly blended for a uniform distribution. A sample of the blended formulation was used to prepare liquid medium by adding 31 g per liter of purified water. The combinations were mixed by hand for about 1 minute to dissolve and autoclaved at 121 °C for 15 minutes. Autoclaving at higher temperatures or for longer periods will result in darkening of the sterile medium, (a darker color does not noticeably affect growth of cells, only the appearance of the sterile medium). |
