Saliva Collection and Preservation Kit

Saliva has been a popular DNA source as an alternative to blood for applications such as genetic studies and pathogen detection. Unlike blood, saliva collection is non-invasive, painless, and easy to perform, and it does not require trained personnel. In addition, saliva does not coagulate, which makes it easier to handle. However, long term storage of saliva samples can be challenging. 
MP Biomedicals’ Preservation Solution rapidly lyses cells and stabilizes DNA within samples. It effectively inhibits the growth of Gram-negative and Gram-positive bacteria and fungi, while also inactivates viruses. This ensures that samples can be safely handled and transported. Furthermore, samples preserved with this solution do not require immediate processing or freezing and can be transported at ambient temperature to centralized testing facilities. The components of the buffer allow samples to be stored for more than 6 months without detectable DNA degradation. Preserved samples can be processed using the SPINeasy DNA Kit for Saliva (Cat. No. 116551050) for DNA extraction. Extracted DNA can be used for various downstream applications including PCR and NGS.

Performance

Figure 1: Performance of four saliva preservation solutions stored at room temperature for 6 months. Genomic DNA was extracted using the SPINeasy DNA Kit Saliva Day 0, 30, 90 and 180. qPCR was performed using 2 μL of gDNA per reaction. Primers targeting bacterial 16S rRNA and human GAPDH were employed. Threshold cycle (Ct) values were compared. Targets were amplified using SYBR Green technology.
