Double dropout formulation (without Histidine and Leucine) of Complete Supplement Mixture (CSM) of amino acids for S. cerevisiae growth media. Time tested formulation is available in complete and dropout formulations.
The selection of plasmids in yeast is based on the use of auxotrophic mutant strains that cannot grow without a specific media component (an amino acid, purine or pyrimidine). Transformation with a plasmid containing the mutated gene enables the transformant to grow on a medium lacking the required component. This CSM-Dropout medium create a richer medium for better yield and growth rate, and increase the probability of successful transformations when screening libraries or performing gene knock-outs.

|pH||pH Range (with DOB, after autoclave): 4.0 - 4.8 at 25 °C|
|Preparation Method||Reagents are tested and chosen for their ability to promote optimum growth of yeast strains when combined in the DOB or DOBA with CSM or CSM-Dropout formula. DOB (Drop Out Base) is 1.7g Yeast Nitrogen Base, 5g Ammonium Sulfate, 20g Dextrose per liter. DOBA is DOB with 17% agar. Chosen reagents are combined in the proper proportions according to the formulation listed in the certificate of analysis and milled to a powder which thoroughly blends the reagents for uniform distribution. A sample of the milled and blended formulation was used to prepare liquid plate medium by adding 43.7g DOBA powder and 0.79g of CSM or the appropriate weight of CSM-Dropout to a liter of purified water. The combinations were mixed by hand for about 1 minute to dissolve the dextrose and autoclaved at 121 °C for 25 minutes. After cooling to 50 °C plates were poured.|
|Product Families Description||Synthetic defined yeast media includes all necessary components for selective growth of Saccharomyces cerevisiae. MP Biomedicals offers synthetic defined broth (SD) and synthetic defined agar (SDA) in a variety of formats for flexibility and convenience. MP Biomedicals offers many amino acid dropout media through combinations of our Complete Supplement Mixture (CSM), several carbon sources, and variations of YNB. If you need amino acid ratios different from those found in our CSM mixtures, we also offer classic amino acid formulations Hollenberg Supplement Mixture (HSM), Brent Supplement Mixture (BSM) and Synthetic Complete/Hopkins Mixture (SC). Synthetic defined media can be ordered in a number of ways: - Order a complete SD or SDA - Order a dropout Base (DOB) or Dropout Base with Agar (DOBA) plus the appropriate amino acid supplement mixture (CSM) - Order a Yeast Nitrogen Base (YNB), a carbon source, the appropriate amino acid supplement mixture (CSM), and agar if desired. - Order all raw materials separately Synthetic Defined Yeast Media: Formulas To Grow By! Synthetic defined yeast media includes all necessary components for selective growth of Saccharomyces cerevisiae. MP Biomedicals offers synthetic defined broth (SD) and synthetic defined agar (SDA) in a variety of formats for flexibility and convenience. Use the following table to navigate through the world of yeast media choices! | SDA = SD recipe + 17 g/L of Agar; DOBA = DOB recipe + 17 g/L of Agar |Use Yeast Nitrogen Base (YNB) Formulations to make your own custom synthetic defined yeast media. Start with the YNB of your choice, add a carbon source, the appropriate amino acids, and agar if necessary. |Dropout Base (DOB) and Dropout Base with Agar (DOBA) is a complete yeast nitrogen base with a carbon source. A complete supplemented synthetic defined medium is easily made by mixing DOB and Complete Supplement Mixture (CSM) which contains amino acids, adenine and uracil. CSM is also availble in Dropout formulations.|Packaging options include: Bulk powder, easily weighed to make custom batch sizes. |Pre-measured, sealed pouches, for 0.5 L or 1 L batch sizes, simply tear open the pouch and add to water. |Pre-poured Plates with agar, Ready-to-Use! |Encapsulated, the Ultimate in Convenience.
