pH Acceptable pH range is 6.9 ± 0.3 at 25 °C. Please note that the pH of CIRCLEGROW™ Medium liquid increases as the temperature of the solution decreases.

Preparation Method Media Preparation: Reagents are tested and chosen for their ability to promote optimum growth of E. coli when combined in Circlegrow™ Agar formulations. Chosen reagents are combined in the accurate proportions according to the proprietary Circlegrow™ Agar formula and thoroughly blended for a uniform distribution.

Product Families Description CIRCLEGROW™ media is designed for obtaining rapid growth of E. coli and high yields of plasmid DNA. Bacteria grown in conventional culture media reach an abrupt stationary phase upon exhaustion of nutrients. Cell proliferation is halted completely and some cell death and plasmid loss occurs. At the end of log phase, cells growing in CIRCLEGROW™ begin to utilize a proprietary second carbon source which becomes available on a delayed basis. Cell growth and plasmid replication continue long after cells in conventional media have reached stationary phase or expired. Bacteria commonly grow in CIRCLEGROW™ to a density triple that achieved in other media. The slow post-log phase growth of bacteria in CIRCLEGROW™ eliminates the need for constant OD monitoring. CIRCLEGROW™ cultures left overnight or over the weekend will have a much lower percentage of dead cells, and a higher percentage of plasmid DNA, than cultures prepared with any other media. The result is a higher yield of plasmid DNA in less volume with less attention.
