Application Notes

Goat F(ab')2 Fragment to Mouse Complement C3 is used as a precipitating antibody. It gives sharp precipitin lines in immunoelectrophoresis and immunodiffusion techniques. It is also suitable for conjugation with enzymes, radiolabels, fluorochromes or other markers, and for attachment to solid supports for use as an immunoadsorbant. Used as a primary reagent in enzyme immunoassays, fluorescein immunoassays and blot, cell or tissue immunostaining. F(ab')2 fragments are recommended for staining of cells or tissues which contain Fc receptors).

Analysis Note The total protein is measured using the Biuret procedure, with bovine albumin as a standard. Antibody titer of the IgG fraction is standardized by equivalence-point precipitation. F(ab')<sub>2</sub> fragments are tested for purity and specificity at a minimum of 20 mg/mL using immunoelectrophoresis; No trace of albumin is detected. Antibody activity to other serum proteins is not present.

Preparation Method The F(ab')<sub>2</sub> fragment is prepared from the specific goat IgG fraction by limited digestion with pepsin, and purified by gel filtration or affinity chromatography. The resulting F(ab')<sub>2</sub> fragment is dialyzed into 0.02M sodium phosphate, 0.14M sodium chloride, 0.05% sodium azide, pH 7.3, adjusted to standard titer, filtered through a 0.22 µm filter, vialed and lyophilized.
