Feeling under water from the difficulty of extracting good quality genomic DNA from marine environment
The isolation of good quality and quantity of DNA from microorganisms in marine sediment can be difficult because of the unique characteristic of the marine microorganisms and the marine environment.
The characteristic of marine microorganisms is interesting and greatly differs from the terrestrial microorganisms in term of both physiological and metabolic features. For example, the marine microorganisms are halophilic and are tolerant to low temperature and high pressure.
At the same time, the access to genomic DNA of microorganisms in marine environment is limited by low biomass turnover and low microbial activity because of extreme living condition and extreme energy limitation, which is scarcely available only as luminous energy. Furthermore, most microorganisms typically stay dormant as endospores.

Sample was prepared in order of protocol in accordance to product guidelines found here.
The researcher adjusted the protocol as recommended, and repeated the DNA extraction with SPINeasy DNA Extraction kit of Soil (Cat No. 116530050), starting with lysis and homogenization steps with FastPrep 24 5G and lysing matrix. This time a good yield and purity was achieved which could meet the downstream experiment requirement.
