A study to examine the ecological role of earthworms and the contents in their gut was stumped by constraints of sample preparation. The DNA extraction results achieved by the researcher, using their kit, was relatively low, with DNA yield of 37.2 ng/μl, the absorbance ratio of 260/280 was 1.796, and the purity was only at 0.044. The yield was insufficient for subsequent amplicon sequencing and high-throughput fluorescent PCR. The researcher would need to repeat the sample preparation to improve on the DNA yield from the unique soil sample comprising of mostly earthworm intestinal content and the surrounding soil in the diet
