Application Notes
3,3',5,5'-Tetramethylbenzidine is suitable as peroxidase substrate for use in ELISA procedures. The substrate produces a soluble end product that is pale blue in color and can be read spectrophotometrically at 370 or 620-650 nm. The TMB reaction may be stopped with 2 M H2SO4 (resulting in a yellow color), and read at 450 nm. A sensitive and specific reagent for the detection of blood, assay of hemoglobin, assay of peroxidases. It is useful in gel staining procedure for low levels of home-associated peroxidases and for enzyme immunoassay of horseradish peroxidase.

| Typical Working Concentration | Solutions for HRP substrate: Dissolve 1.5 mg TMB in 0.1 mL of DMSO. Then add 9.9 ml of 0.1 M sodium acetate (pH 6.0). Finally, add hydrogen peroxide to a concentration of 0.01% and filter through a Whatman #1 paper filter. Prepare solutions fresh daily. Discard any remaining solution at the end of the day. |
