Product Description

FastDNA™ SPIN Kit for Soil

Genomic DNA Extraction from Soil and Environmental Samples

The FastDNA™ SPIN Kit for Soil from MP Biomedicals is designed for genomic DNA extraction from complex soil and environmental samples. The kit combines mechanical bead beating using Lysing Matrix E tubes with chemical processing and silica-based spin-filter purification to support DNA recovery from mixed microbial communities and other biological materials found in environmental matrices.

This integrated workflow helps researchers prepare genomic DNA for downstream molecular biology applications, including PCR-based analysis, microbial detection, and sequencing workflows where sample quality and assay requirements have been validated.

How the Workflow Works

Soil samples are mechanically disrupted using a compatible FastPrep instrument and the supplied Lysing Matrix E tubes. Subsequent precipitation, DNA binding, washing, and elution steps help separate nucleic acids from cellular debris and environmental contaminants, including PCR-inhibitory compounds.

Processing parameters and reagent preparation should follow the current kit instructions and compatible instrument operating guidelines.

Research Applications

  • Soil Microbiology: Extraction of genomic DNA for microbial community analysis and environmental microbiology research.
  • Environmental DNA Research: DNA extraction from soil, sediment, and related environmental sample matrices.
  • PCR-Based Molecular Analysis: Preparation of genomic DNA for microbial detection, amplification, and molecular research.
  • Sequencing Workflows: Sample preparation for downstream sequencing applications where DNA quality and assay requirements have been validated.

Frequently Asked Questions

1. What is the FastDNA SPIN Kit for Soil used for?

The FastDNA SPIN Kit for Soil is used to extract genomic DNA from soil and other environmental samples, including mixed microbial populations. The workflow combines FastPrep bead-beating lysis with silica-based DNA purification for downstream molecular research.

2. How much soil can be processed per extraction?

The standard 2 mL FastDNA SPIN Kit for Soil workflow accepts up to 500 mg of soil per preparation. Follow the current manual when working with unusual or highly inhibitory samples.

3. Does the kit include Lysing Matrix tubes?

Yes. The standard 50-preparation kit includes 50 Lysing Matrix E tubes (2 mL) for mechanical disruption using a compatible FastPrep instrument.

4. Is a FastPrep instrument required?

The validated workflow is designed for mechanical homogenization with a FastPrep instrument and Lysing Matrix E tubes. Confirm the supported instrument and adapter configuration before use.

5. How does the kit address PCR inhibitors in soil?

Following lysis, the protocol uses precipitation, silica-based DNA binding, and wash steps to help remove humic acids and other contaminants that can interfere with PCR. Removal efficiency can vary with the sample matrix.

6. Which environmental samples can be used?

The kit is intended for soil and other environmental materials such as sediments, sludge, compost, and rhizosphere-associated samples. Sample-dependent optimization may be needed.

7. Is the extracted DNA suitable for PCR?

The kit is designed to yield DNA suitable for PCR and other downstream molecular applications. Suitability for a particular assay or sequencing workflow should be evaluated using the resulting DNA quality and relevant controls.

8. What is the difference between FastDNA SPIN Kit for Soil and SPINeasy DNA Pro Kit for Soil?

Both are MP Biomedicals kits for soil DNA extraction. FastDNA SPIN Kit for Soil uses Lysing Matrix E and a silica-based spin-filter workflow, whereas SPINeasy DNA Pro Kit for Soil uses its own kit-specific reagents and Lysing Matrix YB. Select based on sample type, workflow, and downstream requirements.
