The SPINeasy® DNA Kit for Forensics uses advanced silica-membrane spin-column technology to deliver high-performance DNA extraction from a wide range of forensic casework samples. It provides strong overall DNA recovery, including genomic and mitochondrial DNA, with no inhibition in downstream applications.
The protocol is simple, requires no specialized equipment, and is formulated to effectively lyse and extract DNA from challenging forensic materials such as stains, swabs, adhesive-backed samples, nail clippings, teeth, chewing gum, cigarette butts, and small volumes of blood or saliva.
The kit’s purification system ensures selective DNA binding and high-quality recovery suitable for immediate use in downstream applications, including PCR and STR analysis. Its reliable performance supports high success rates in STR profiling, making it ideal for maximizing DNA recovery from limited or critical forensic samples.

Figure 1: Yield of DNA Extracted from simulated casework samples.
Sample types: Fingerprint tape (fingerprint collected on the sticky side of tape, 50 cm²); Cigarette butt (outer paper from the end of the cigarette, 1 cm²); Hair with follicle (three strands cut 1 cm from the hair bulb); Blood stain (5 μL of blood applied to 1 cm² filter paper); Nail clipping (10 mg minced); Tooth (10 mg of powdered tooth); Chewing gum (~100 mg); Oral rinse (100 μL collected after a 60-second rinse with sterile saline); Oral swab (buccal cells collected by rolling the swab firmly 10 times on each cheek); Blood (50 μL of whole blood). Blue bars represent the SPINeasy® DNA Kit for Forensics (MP), and grey bars represent Kit Q.

Figure 2: Amplifiability of DNA extracted with SPINeasy® DNA Kit for Forensics
Ct values of real time PCR using DNA isolated from various sample types using SPINeasy® DNA Kit for Forensics and Kit Q. Left: Ct values obtained using primers targeting CSF1PO (STR locus on chromosome 5). Middle: Ct values obtained using primers targeting DXS10079 (STR locus on X chromosome). Right: Ct values obtained using primers targeting Human mitochondrial genome NC_012920 (hmito).

Figure 3: Long fragment PCR using DNA extracted with SPINeasy® DNA Kit for Forensics
Agarose gel electrophoresis images showing PCR amplification products of the partial mitochondrial CO1 gene (827 bp). DNA templates were extracted from five different sample types using the SPINeasy DNA Kit for Forensics and Kit Q.
MP: Tooth from single female donor

Kit Q: Tooth from single donor (female)

MP: Blood stain pooled whole blood from multiple donor
Kit Q: Blood stain pooled whole blood from multiple donor
Figure 4: STR analysis using DNA extracted with SPINeasy® DNA Kit for Forensics and Kit Q
STR analysis results showing genotyping results on 30 STR loci. The DNA samples used were extracted with SPINeasy® DNA Kit for Forensics and Kit Q from two different sample types, a tooth from a female donor (A and B) and blood stain which contains blood from multiple donor (C and D).
When comparing DNA samples extracted from Blood stain using the 2 Kits, the STR analysis results were highly similar. Since the Blood stain sample were originated from multiple donors, multiple peaks were identified at most of the loci. There were no difference in number of peaks identified using the two DNA isolation kits.
