Product Description

Ficoll® is a synthetic, hydrophilic polymer made by the copolymerization of sucrose and epichlorhydrin. It is a highly branched polymer. Solutions at neutral pH can be sterilized by autoclaving. Ficoll® shows following key Advantages:

•Proven medium for density gradient centrifugation.
•Does not penetrate biological membranes
•High density solution of low viscosity and low osmotic pressure
•Preserves functional and morphological integrity.
•Useful for separating cells which are sensitive to centrifugation and for separating cells of similar density but different size (under conditions of sedimentation at unit gravity).
•Can be used in other applications such as electrophoresis, hybridization cryopreservation and as a hapten carrier.

Application Notes

Ficoll® is widely used as a density gradient centrifugation medium. Solutions of Ficoll® containing up to 50% (w/v) can be obtained to cover a density range up to 1.2 g/mL. Ficoll® has a much lower osmotic pressure than equivalent concentrations of sucrose thus allowing construction of isotonic density gradients and preservation of physiological and morphological integrity during centrifugation. It is used for the routine isolation of blood cells and for separation of cells and organelles by unit gravity sedimentation. It is also very suitable as an immunologically inert carrier for low molecular weight haptens in immunological studies. This is commonly used to reduce nonspecific binding of material to nitrocellulose membranes during nucleic acid hybridization experiments. Loading nucleic acids onto agarose gels for electrophoresis is easier in the presence of Ficoll®.
