Product Description

2-Macroglobulin is a tetrameric glycoprotein which acts as a broad-range irreversible protease inhibitor. It is found abundantly in plasma and interstitial fluids. The protease:alpha2-Macroglobulin balance plays an important role in mediating inflammation-associated tissue destruction. Serum levels of alpha2-macroglobulin and protease:alpha2-macroglobulin complexes are increased in patients with sepsis, emphysema, peridontitis, rheumatoid arthritis, and other inflammatory diseases. It is possible that the oxidant inactivation of alpha2-macroglobulin contributes to tissue destruction in inflammation. alpha2-Macroglobulin has been implicated as a genetic risk factor for late-onset Alzheimer's disease. Activated alpha2-macroglobulin enhances the clearance of soluble alpha/beta-amyloid via low-density lipoprotein receptor-related protein in cortical neurons but has no effect on secredted or full-length APP levels.

When the protease cleaves the macroglobulin "bait" sequence, the macroglobulin rearranges and traps the protease. It inhibits bromelain, chymotrypsin, elastase, endoproteinase Arg-C, endoproteinase Glu-C, ficin, papain, plasmin, subtilisin, thermolysin, thrombin, and trypsin. It is known to inhibit neurite outgrowth by blocking the activity of b-nerve growth factor.

Biochemical Physiological Actions One mg of α2-macroglobulin will inhibit a minimum of 10 μg trypsin with an activity of 10,000 BAEE un/mg protein. α2-Macroglobulin is found abundantly in plasma and interstitial fluids. The protease-α2-M balance plays an important role in mediating inflammatory tissue destruction. Serum levels of α2-M and protease-α2-M complexes are increased in patients with sepsis, emphysema, periodontitis, rheumatoid arthritis, and other inflammatory diseases, and oxidant inactivation of α2-M may contribute to tissue destruction during inflammation.
