This product is made by a modification of Freund's technique, in which of heat-killed Mycobacterium tuberculosis (0.05%) is suspended in a mixture of paraffin oil (85%) and Arlacel A (15%). Freund's Complete Adjuvant has been widely used as a potent adjuvant in immunization protocols and vaccine development.
The Mycobacterium tuberculosis in Freund's Complete Adjuvant contains a unique glycolipid trehalose 6,6’ dimycolate (TDM, also known as cord factor), which stimulates macrophage-inducible C-type lectin (Mincle). Additionally, FCA has ligands for TLR2, TLR4, and TLR9.
The oil emulsion in Freund's Complete Adjuvant helps to retain the antigen at the injection site, providing a slow release of the antigen over an extended period, thus facilitating a prolonged and robust immune response. The oil emulsion also triggers the activation of complement and other effector molecules that enhance the clearance of the antigen and the recruitment of immune cells to the injection site.
Freund's Complete Adjuvant is primarily used as an initial injection in immunization protocols, as subsequent injections with Freund's Complete Adjuvant are generally avoided due to the risk of inducing adverse reactions.
