链脲佐菌素(Streptozocin,STZ)是一种氨基葡萄糖—亚硝基脲衍生物,具有促甲基化、致癌、抗菌并诱导糖尿病的特性。相对于其他诱导试剂因其对机体组织毒性相对较小,动物存活率高,是目前国内外糖尿病造模使用最多的一种方法。

STZ诱导I型糖尿病

方法:         I型糖尿病显著的病理生理学特征为胰岛β细胞数量显著减少和消失所导致的胰岛素分泌显著下降或缺失。采用较大剂量一次性注射的给药方法,可直接引起胰岛β细胞的广泛破坏,造成Ⅰ型糖尿病模型。

优势:         方法简便,成模时间短,成模率高,与人类I型糖尿病表现非常相似。

高脂饲料+STZ诱导Ⅱ型糖尿

方法:         Ⅱ型糖尿病显著的病理生理学特征为胰岛素调控葡萄糖代谢能力的下降(胰岛素抵抗)伴随胰岛β细胞功能缺陷所导致的胰岛素分泌减少(或相对减少)采用高糖高脂饲养并配合低剂量注射的方法,可建立Ⅱ型糖尿模型。胰岛素抵抗可以通过高糖、高脂饮食实现。而胰岛细胞功能缺陷引起的胰岛素分泌减少一般采用STZ诱导。

优势:         实验周期短、方法简便、相对可靠稳定、造模成本低、且诱导的Ⅱ型糖尿病模型症状和发病机制与人类Ⅱ型糖尿病非常相似。

Typical Working Concentration Has been used in intravenous injections in rats at a dose of 65 mg/kg body weight to induce diabetes (using cold 0.1 M citrate buffer pH 4.5). In rats and dogs, diabetes was induced using intravenous dosage of 50 mg/kg body weight (using 1-2% w/v solutions in saline buffered with citrate dextrose solution at pH 5.0).
