YES Agar is a nutritionally rich medium for the growth of S. pombe, containing dextrose, yeast extract and agar. Dextrose is the energy source. Yeast extract provides carbon, nitrogen, minerals, vitamins, trace ingredients and other essential growth nutrients which stimulate the growth of yeasts.
EMM (Edinburgh Minimal Medium) is used for the growth of fission yeast S. Pombe.

|Preparation Method||Media Preparation: Reagents are tested and chosen for their ability to promote optimum growth of S. pombe when combined in YES Agar formulations. Chosen reagents are combined in the proper proportions according to the YES Agar formulation and thoroughly blended for a uniform distribution. A sample of the blended formulation is used to prepare agar medium. The combinations are mixed to dissolve components and then autoclaved at 121 °C for 15 minutes.|
