SuperBroth is enriched medium with high levels of tryptone and yeast extract. Due to the high levels of peptides, amino acids, vitamins and cofactors high yields of E. coli cells and lambda bacteriophages are obtained.

Preparation Method | Media Preparation: Reagents are tested and chosen for their ability to promote optimum growth of lambda phage and phagemids in E. coli when combined in SuperBroth formulations. Chosen reagents are combined in the proper proportions according to the Superbroth formulation and thoroughly blended for a uniform distribution. A sample of the blended formulation was used to prepare liquid medium by adding 60 g per liter of purified water. The combinations were mixed by hand for about 1 minute to dissolve and autoclaved at 121 °C for 15 minutes. Autoclaving at higher temperatures or for longer periods will result in darkening of the sterile medium, (a darker color does not noticeably affect growth of cells, only the appearance of the sterile medium). |
