Application Notes

Alkaline phosphatase-conjugated rabbit affinity purified antibody to goat IgG (whole molecule) is suitable for use in immunoblotting. The product can also be used for direct ELISA (1:30,000) and immunohistochemistry (1:50 using formalin-fixed, paraffin-embedded sections). Detection of TLR2 and TLR4 in protein extracts from epidermal keratinocytes was performed by western blot using alkaline phosphatase conjugated rabbit anti-goat IgG as the secondary at a 1:2500 dilution. Polyclonal anti-goat IgG (whole molecule)–alkaline phosphatase antibody can be used in immunoblotting and ELISA. Immunostaining of acetone-fixed frozen and formalin-fixed, paraffin-embedded tissue sections.

Preparation Method Antibody and highly purified calf-intestine alkaline phosphatase are conjugated under defined conditions to obtain optimally labeled product. Conjugated protein is purified by gel filtration. The product is dialyzed into 0.01M TRIS-HCl, 0.25M sodium chloride, pH 8.0, with 15 mg/mL bovine albumin and 0.05% sodium azide, adjusted to standard titer, filtered through a 0.22 μm filter, and vialed.
