Superior Extraction and Purification

The MPure-12™ System employs an advanced magnetic bead separation technology that enables rapid and efficient purification of nucleic acids resulting in the highest yields and purity. This process includes four main steps: lysis, binding, washing and elution. Purifying the nucleic acids with the MPure-12™ System takes only 35 to 70 minutes depending on the selected protocol and kit. MPure-12™ Reagent Kits

MPure-12™ nucleic acid purification reagent kits used with the MPure-12™ system provide the highest yield and purity. MP Biomedicals offers 15 different MPure-12™ reagent kits that allow for the purification of DNA and RNA from a wide range of bio-specimens. MPure-12™ kits contain everything needed for sample purification: Pre-filled reagent cartridges, reaction chambers, sample tubes, elution tubes, disposable pipette tips, etc.

MPure-12™ pre-filled reagent cartridges are designed to increase yield, purity and data quality while decreasing contamination risk and cost. Reagent cartridges are pre-measured and film-sealed to avoid contamination; the MPure-12™ System automatically pierces this film before moving the reagent to the sample tube, eliminating operator handling errors. Each cartridge contains reagents for one sample and cartridges may be run individually, allowing flexibility in the number of samples processed (from 1 to 12) with no unnecessary waste of reagents.

Integrity

The DNA was isolated in replicates of 12 from 200 -L human whole blood taken from two different donors. Elution volume was set to 100 -L. Integrity of DNA was shown by subjecting each eluate to TAE agarose gel electrophoresis together with a suitable molecular weight marker. All samples exhibit single band with molecular weight > 40 kb with no smear.

Reproducibility

The DNA was isolated by MPure-12™ blood DNA extraction kit from twenty whole blood samples. The --globin gene was detected by real-time qPCR. This data shows ultra high stability and reproducibility of MPure-12™-s nucleic acid purification capability.

The DNA was extracted from whole blood by MPure-12™ Blood DNA extraction kit 200. Real-time qPCR detection of the --globin (A) and spiked parvovirus B-19 DNA (A,B). No significant influences on samples with different anticoagulants and storage conditions.

Sensitivity

Whole blood (in EDTA collection tube) was spiked with serial-dilution of human Parvo B19 Virus (in the range of 5-500,000 IU/L). 200 -L samples were extracted and eluted in 100 -L volume. 10 -L of eluate was used for real-time PCR reaction by RealStar- Parvovirus B19 PCR kit 1.0. As little as 5 IU spiked (about 1 IU in PCR reaction) sample can be detected, proving the excellent sensitivity and linearity of isolation.

The DNA was isolated from different whole blood samples (WBCs count range 1.8 - 22 x 103 cells/-L). DNA yield (measured by Nanodrop 2000) shows excellent scalability in different volumes (100, 200, 300 and 400 -L) of extraction and WBCs count of the samples.
