Isolation of genomic DNA from 100 mg of Andisol samples using the FastDNA™ Spin Kit for Soil. Samples were processed (40 sec at 6 m/s) using a FastPrep-24™ instrument. Extracted genomic DNA (left = top) and PCR products (right = bottom) from equivalent amounts of andisol samples (1-7) were analyzed in agarose/ ethidium bromide gel. M is a DNA size marker.

Agarose gel electrophoresis of DNA and PCR products extracted from sample soils (1-7). FastPrep was used for extracting DNA from soils (A). PCR products from these extracts were amplified with bacterial 16S rDNA universal primer set (338f and 907r) (B) when amended with (+) or without (-) 40 mg of skim milk g-1 soil. Numbers show soil samples listed in Table 1. M: Molecular marker (A: λ/Hind III digest, B: 100 bp ladder).
The combination of the FastPrep® instrument and the FastDNA™ SPIN Kit for soil have demonstrated successful extraction of PCR-suitable DNA from recalcitrant soil sample.

Soil types differ in the type and amount of organic materials, and the largest and most chemically significant fraction of natural organic matter is the humic substances.

The amount and type of humic substances in a soil sample are established by a combination of environmental conditions, vegetation and topography. Humic substances frequently give soil a yellow/brown color and have been shown to inhibit Taq polymerase activity at concentration as low as 0.1 mg/ml.

FastRNA™ Pro Soil Kits purify RNA in a process that removes humic substances and other inhibitors, and efficiently inactivates cellular Rnases during homogenization to prevent RNA degradation.

Each kit offers two levels of RNA purification that permit tailoring the protocol to the soil sample and downstream applications.
